RESUMO

The contribution of branched-axon monosynaptic inputs in the generation of short-term synchronization of motoneurones remains uncertain. Here, synchronization was measured for intercostal and abdominal motoneurones supplying the lower thorax and upper abdomen, mostly showing expiratory discharges. Synchronization in the anaesthetized cat, where the motoneurones receive a strong direct descending drive, is compared with that in anaesthetized or decerebrate rats, where the direct descending drive is much weaker. In the cat, some examples could be explained by branched-axon monosynaptic inputs, but many others could not, by virtue of peaks in cross-correlation histograms whose widths (relatively wide) and timing indicated common inputs with more complex linkages, e.g., disynaptic excitatory. In contrast, in the rat, correlations for pairs of internal intercostal nerves were dominated by very narrow peaks, indicative of branched-axon monosynaptic inputs. However, the presence of activity in both inspiration and expiration in many of the nerves allowed additional synchronization measurements between internal and external intercostal nerves. Time courses of synchronization for these often consisted of combinations of peaks and troughs, which have never been previously described for motoneurone synchronization and which we interpret as indicating combinations of inputs, excitation of one group of motoneurones being common with either excitation or inhibition of the other. Significant species differences in the circuits controlling the motoneurones are indicated, but in both cases, the roles of spinal interneurones are emphasised. The results demonstrate the potential of motoneurone synchronization for investigating inhibition and have important general implications for the interpretation of neural connectivity measurements by cross-correlation.

The conventional method for quantification of polyhydroxyalkanoates based on whole-cell methanolysis and gas chromatography (GC) is laborious and time-consuming. In this work, a method based on flow cytometry of Nile red stained bacterial cells was established to quantify poly-3-hydroxybutyrate (PHB) production by the diazotrophic and plant-associated bacteria, Herbaspirillum seropedicae and Azospirillum brasilense. The method consists of three steps: i) cell permeabilization, ii) Nile red staining, and iii) analysis by flow cytometry. The method was optimized step-by-step and can be carried out in less than 5 min. The final results indicated a high correlation coefficient (R2=0.99) compared to a standard method based on methanolysis and GC. This method was successfully applied to the quantification of PHB in epiphytic bacteria isolated from rice roots.

In contrast to climacteric fruits, in which ethylene is known to be pivotal, the regulation of ripening in non-climacteric fruits is not well understood. The strawberry is a typical example of a non-climacteric fruit, which has been used as a model system of these types of fruit. In this study, the effect of exogenous ethephon on the expression of ethylene biosynthesis and signaling genes, FaERF2 and FaACO1, was analyzed in the Fragaria ananassa cultivar Camino Real by quantitative real-time polymerase chain reaction, and the physicochemical and phytochemical characteristics of fruits were evaluated in field trials and postharvest tests. Transcript accumulation was influenced by exogenous treatment with ethephon, which affected the pattern of gene expression during different stages of growth and fruit development, with the highest expression occurring during postharvest tests. In addition, ethephon significantly influenced the phytochemical profile of sugars, anthocyanins, phenolic compounds, and vitamin C contents both in the field- and postharvest-treated fruits at different stages. These results indicate that ethylene regulates the phenylpropanoid maturation pathway in strawberry fruit.

RESUMO Objetivou-se avaliar o potencial alelopático de extratos aquosos de braquiária, girassol e sorgo na germinabilidade e crescimento inicial de alface (Lactuca sativa L.) visando o uso destes extratos como herbicida natural. O estudo foi realizado no Laboratório de Fisiologia Vegetal da Universidade Federal do Recôncavo da Bahia. Foi instalado um experimento no delineamento inteiramente casualizado, com 4 tratamentos e 6 repetições. Os tratamentos foram extratos de braquiária, girassol, sorgo e como controle, água destilada. Os extratos foram obtidos, utilizando 200 g de material vegetal para 1000 mL de água destilada. Os aquênios de alface foram distribuídos em caixa gerbox contendo duas folhas de papel germitest umedecidas com os tratamentos e levadas para câmara de germinação. A germinação foi acompanhada diariamente. Foram avaliados o tempo médio de germinação e, após dez dias, o crescimento inicial, medindo-se o comprimento da parte aérea (CPA) e o comprimento do sistema radicular (CSR). Os dados gerados foram submetidos à análise de variância e as médias comparadas pelo teste de Tukey. Com base nos resultados, os extratos de braquiária, girassol e sorgo não afetaram a germinação da alface, mas os de braquiária e sorgo aumentaram o tempo médio de germinação. Os extratos causaram aumento na parte aérea, diminuíram o crescimento do sistema radicular e a formação de plântulas normais. Por causarem inibição no crescimento do sistema radicular da alface, apresentam potencial para serem utilizados como herbicidas naturais.


ABSTRACT This study aimed to evaluate the allelopathic potential of aqueous extracts of Brachiaria, sunflower, sorghum germination and early growth of lettuce (Lactuca sativa L.), in the attempt of employing these extracts as a natural herbicide. The study was conducted at the Laboratory of Plant Physiology of the Universidade Federal do Recôncavo da Bahia. A trial was arranged in a completely randomized design with 4 treatments and 6 replications. The treatments were extracts of Brachiaria, sunflower, sorghum, and, as a control, distilled water. The extracts were obtained by using 200 g of plant material in 1000 mL of distilled water. First, the lettuce seeds were distributed in a seedling box containing two sheets of germitest moistened paper with treatments, and then were taken to a germination chamber. The germination was daily monitored. The average germination time and the initial growth were evaluated after ten days, through the measuring of the aerial part length (APL) and the root system length (RSL). The data generated were subjected to analysis of variance and the means were compared by the Tukey test. Based on the results, the extracts of Brachiaria, sunflower and sorghum did not affect the germination of lettuce, but, on the other hand, the Brachiaria and sorghum increased the germination time. The extracts caused an increase in the aerial part and reduced the root system growth and the formation of normal seedlings. Since they caused inhibition on the growth of the lettuce root system, have also potential to be used as natural herbicides.

RESUMO

Forty Santa Ines sheep (Ovis aries Lin.) hearts were examined with the purpose of studying the present types of chordae  tendineae in the left valve complex. The study was performed at the State University of North Fluminense Darcy Ribeiro, Morphology sector of the Agricultural Science Animal Health Center Laboratory. Hearts were initially kept in 10% formalin solution and later dissected to undergo removal and plannifcation of left atrioventricular complex. With the removal of the left atrioventricular complex began the observation and characterization of the cord. After the analysis we observed the presence of eight types of cuspidal chordae tendineae present in the left valve complex, they were classifed as type I, II, III, IV, V, VI, VII and commissural kind.

Hydrophobic films of polystyrene synthesized in bulk (PS) and by emulsion polymerization in the presence of the cationic surfactant cetyltrimethylammonium bromide (PS-CTAB) or the anionic surfactant sodium dodecyl sulfate (PS-SDS) were characterized by means of ellipsometry, contact angle measurements, and atomic force microscopy. Thin (approximately 65 nm) and thick (approximately 300 nm) films were spin-coated on hydrophilic silicon wafers. PS films presented scarcely tiny holes, while PS-CTAB and PS-SDS films presented holes and protuberances. The former were attributed to dewetting effects and the latter to surfactant clusters. The films were exposed to water or to a 0.1 mol/L NaCl solution for 24 h. Ex situ measurements evidenced strong topographic alterations after the exposure to the fluid. A model based on the diffusion of water (or electrolyte) molecules to the polymer/silcon dioxide interface through holes or defects on the film edges was proposed to explain the appearance of wrinkles and protuberances. In situ ellipsometric measurements were performed and compared with simulations, which considered either a water layer between a polymer and a silcon dioxide layer or an air layer between a polymer and water (medium). In the case of thin PS films, the ellipsometric angles evidenced a very thin (0.5-1.0 nm) air layer between water and the PS films. Upon increasing the PS film thickness, no air layer could be observed by ellipsometry. Regardless of the thickness, the ellipsometric data obtained for PS-CTAB and PS-SDS films did not indicate the presence of an air layer between them and the aqueous media. The dramatic changes in the topography of PS, PS-CTAB, and PS-SDS after immersion in salt solution were explained with proposed models. From a practical point of view, this study is particularly relevant because many hydrophobic polymers are used as substrates for biomedical purposes, where the physiological ionic strength is 0.15 mol/L NaCl.

The biological cycle of female ticks of Amblyomma cooperi was studied under controlled conditions. The study has begun with two engorged females collected in a naturally infested wild capybara. The larvae originated from the oviposition of these two females were used for collecting young adults through artificial infestations in rabbits. The female parasitic and free living phases were evaluated using artificial infestation of ticks in a capybara. The average body weight of female was 958.2+/-175.6mg, the average pre-egg laying period was 8.5+/-1.4 days, the reproductive efficiency index was 59.5+/-4.2 and the nutritional efficiency index was 77.3+/-4.8. The incubation and eclosion periods were 41.0 and 5.0 days, respectively, and the rate of eclosion was 64 per cent. These figures were obtained under hight relative humidity conditions which were necessary for the sucess of the egg incubation process of this species. The preliminary data obtained with the artificial infestation in rabbits raises the possibility of this and other domestic species be used as an epidemiologic link between the domestic and the wild environment with the potential exposure of human populations to A. cooperi and to the maculosa fever agent

In this paper, we address the problem of navigating an autonomous mobile robot in an unknown indoor environment. The parti-game multiresolution learning approach is applied for simultaneous and cooperative construction of a world model, and learning to navigate through an obstacle-free path from a starting position to a known goal region. The paper introduces a new approach, based on the application of the fuzzy ART neural architecture, for on-line map building from actual sensor data. This method is then integrated, as a complement, on the parti-game world model, allowing the system to make a more efficient use of collected sensor information. Then, a predictive on-line trajectory filtering method, is introduced in the learning approach. Instead of having a mechanical device moving to search the world, the idea is to have the system analyzing trajectories in a predictive mode, by taking advantage of the improved world model. The real robot will only move to try trajectories that have been predicted to be successful, allowing lower exploration costs. This results in an overall improved new method for goal-oriented navigation. It is assumed that the robot knows its own current world location-a simple dead-reckoning method is used for localization in our experiments. It is also assumed that the robot is able to perform sensor-based obstacle detection (not avoidance) and straight-line motions. Results of experiments with a real Nomad 200 mobile robot are presented, demonstrating the effectiveness of the discussed methods.
